Eukaryotic transcription is most often under positive regulation: A transcription factor is provided under tissue-specific control to activate a promoter or set of promoters that contain a common target sequence.

The PCR primers must not have self complementary regions as this would lead to hairpin formation within primer molecules and make then unsuitable for PCR.

Proteins differ in the amount of hydrophobic amino acids that are present on their surface. This difference can be exploited in salt fractionation but can also be used in a higher resolution method using hydrophobic interaction chromatography (HIC).

#Unit 3. Fundamental Processes

Sites of phosphorylation on the CTD serve as a recognition or anchor point for other proteins to dock with the polymerase. The capping enzyme (guanylyl transferase), which adds the G residue to the 5' end of newly synthesized mRNA, binds to CTD phosphorylated at serine 5, the first phosphorylation event catalyzed by TFIIH.
P-TEFb leads to recruitment of a set of proteins called SCAFs to the CTD, and they in turn bind to splicing factors. This may be a means of coordinating transcription and splicing.
